Prodrug মানে কী?
ড্রাগের এমন একটা form যেটা মূলত inactive, কিন্তু আমাদের বডিতে administration এর পর active হবে।
Prodrug কেনো দেয়া হয়?কোনো লাভ আছে কী সেখানে?
হ্যা, অবশ্যই আছে!
-অনেক ক্ষেত্রে active form এ absorption কম হয়,কিন্তু inactive form এ absorption বেশি হয় ড্রাগের।absorption বাড়াতে দেয়া হতে পারে।
-ড্রাগ যেনো active হবার আগে first pass metabolism তথা liver, intestine এ metabolized হয়ে excreted না হয়,এটা কমানোর জন্যও prodrug দেয়া হয়।
-active drug receptor এ যাবার আগে কিছু লোকাল adverse effect করতে পারে,অনেকসময় prodrug use করে সেই adverse effect কমানো হয়।
-bioavailability বাড়ানোর জন্য তথা systemic circulation এ বেশি যেনো active drug আসে সেটা নিশ্চিত করতেও prodrug দেয়া হয়।
-আবার route of administration কে মডিফাই করতেও দেয়া হয়।যেমন :phenytoin -antiepileptic drug এটা নরমালি parenteral route এ দেয়া যায়না।কিন্তু আমরা এটাকে inactive prodrug হিসেবে (fosphenytoin) এ রুপান্তরিত করে parenteral route এ দিতে পারি,এবং এটা plasma এ যাওয়া মাত্রই phenytoin এ convert হবে!
-offensive odor দূর করতেও দেয়া হয়
কিছু prodrug এর example :
1.Levodopa >এটা dopamine এর prodrug.
2.prednisone > এটা prednisolone এর prodrug.
